چکیده
Surface immobilization and characterization of the functional activity fibronectin (Fn) type-III domains are reported. The FnIII9-10 or FnIII10 containing RGD loop PHSRN synergy site were recombinantly produced covalently bound to chemically activated PEG methacrylate (MA) hydrogel coatings by microcontact printing. Such fabricated biochip surfaces 6 mm in diameter consisted 190 µm wide protein stripes separated 200 spacing. They analyzed imaging null ellipsometry, atomic force microscopy fluorescence microscopy. Also, tested human foreskin fibroblast HeLa cultures for at least 96 h, thus evaluating their suitability controlled cell adhesion proliferation. However, while equally well responsive FnIII9-10, Fn surfaces, fibroblasts displayed lower focal areas, as proliferation rates on fragment compared Fn. Nevertheless, full was confirmed immunostaining cells adherent surfaces. observed interaction differences that either type surface composition-dependent demonstrate potential use specifically engineered other ECM protein-derived architectures.

Fibronectin adsorption on hydrophilic and hydrophobic surfaces detected by antibody binding and analyzed during cell adhesion in serum-containing medium.
Studies have been carried out on the adsorption properties of plasma fibronectin (FN) on hydrophobic and hydrophilic surfaces. The hydrophobic and hydrophilic surfaces used were nonwettable bacteriological dishes and wettable tissue culture dishes, respectively. At low plasma FN concentrations, there was more plasma FN binding to hydrophobic surfaces than to hydrophilic surfaces. Under these co...
متن کاملCrystal structures of fibronectin-binding sites from Staphylococcus aureus FnBPA in complex with fibronectin domains.
Staphylococcus aureus can adhere to and invade endothelial cells by binding to the human protein fibronectin (Fn). FnBPA and FnBPB, cell wall-attached proteins from S. aureus, have multiple, intrinsically disordered, high-affinity binding repeats (FnBRs) for Fn. Here, 30 years after the first report of S. aureus/Fn interactions, we present four crystal structures that together comprise the stru...
متن کاملCell-type specific recognition of RGD- and non-RGD-containing cell binding domains in fibrillin-1.
The fibrillins are large glycoprotein components of 10-nm microfibrils found in the extracellular matrix of most tissues. Microfibrils play a role in elastic fiber assembly and serve to link cells to elastic fibers in the extracellular matrix. To determine whether fibrillin-1 specifically interacts with receptors on cells from fibrillin-rich tissues, we evaluated whether two cell types that pro...
متن کاملComparative cell response to artificial extracellular matrix proteins containing the RGD and CS5 cell-binding domains.
This study addresses endothelial cell adhesion and spreading on a family of artificial extracellular matrix (aECM) proteins designed for application in small-diameter vascular grafts. The aECM proteins contain domains derived from elastin and from fibronectin. aECM 1 contains the RGD sequence from the tenth type III domain of fibronectin; aECM 3 contains the fibronectin CS5 cell-binding domain....
متن کاملPolygonal tiling of some surfaces containing fullerene molecules
A tiling of a surface is a decomposition of the surface into pieces, i.e. tiles, which cover it without gaps or overlaps. In this paper some special polygonal tiling of sphere, ellipsoid, cylinder, and torus as the most abundant shapes of fullerenes are investigated.
